WE HAVE ESTABLISHED THE COOPERATIVE INSTITUTE FOR CLIMATE, OCEAN, AND ECOSYSTEM STUDIES (CICOES), A CO-LOCATED, CONSORTIUM COOPERATIVE INSTITUTE THAT INCLUDES THE UNIVERSITY OF WASHINGTON, THE UNIVERSITY OF ALASKA FAIRBANKS, AND THE COLLEGE OF EARTH, OCEAN, AND ATMOSPHERIC SCIENCES AT OREGON STATE UNIVERSITY. THE CICOES RESEARCH INSTITUTE CO-LOCATES CICOES STAFF AT NOAA PARTNER LABORATORIES INCLUDING THE PACIFIC MARINE ENVIRONMENTAL LABORATORY (PMEL), THE ALASKA FISHERIES SCIENCE CENTER (AFSC), AND THE NORTHWEST FISHERIES SCIENCE CENTER (NWFSC). THE CONSORTIUM COMPONENT OF CICOES INCLUDES AFFILIATED FACULTY AND STAFF AT ALL PARTNER UNIVERSITIES WHO CONTRIBUTE INTELLECTUAL EXPERTISE, RESEARCH CAPACITY, TECHNOLOGICAL DEVELOPMENT, EDUCATION AND TRAINING OF POTENTIAL NEXT-GENERATION NOAA EMPLOYEES, AND CONDUCT PUBLIC EDUCATION AND OUTREACH. THE GEOGRAPHIC RANGE OF CICOES EXTENDS FROM THE OREGON-CALIFORNIA BORDER, THROUGH WASHINGTON STATE TO THE GULF OF ALASKA, BERING SEA, CHUKCHI SEA, AND BEAUFORT SEA. ADDITIONAL RESEARCH EFFORTS ARE CONDUCTED IN THE SOUTHERN AND EQUATORIAL REGIONS OF THE PACIFIC OCEAN AND IN THE ATLANTIC, INDIAN, AND SOUTHERN OCEANS. RESEARCH PROGRAMS AND PROJECTS DETAILED IN THIS PROPOSAL ILLUSTRATE THE CAPABILITY, CAPACITY, AND VISION OF CICOES AND THE ALIGNMENT WITH NOAAS MISSION, RESEARCH, AND EDUCATION OBJECTIVES. ARCH REACH: ARCH CENTER FOR RESEARCH AT THE INTERSECTION OF EQUITY, ALCOHOL, COMMUNITY AND HIV - ABSTRACT THE ALCOHOL RESEARCH CONSORTIUM IN HIV CENTER FOR RESEARCH AT THE INTERSECTION OF EQUITY, ALCOHOL, COMMUNITY AND HIV (ARCH REACH) SEEKS TO OPTIMIZE WELL-BEING AMONG PWH AND ACCELERATE PROGRESS TOWARDS ENDING THE HIV EPIDEMIC (EHE) BY ADDRESSING CO-OCCURRING UNHEALTHY ALCOHOL USE (UAU). GROUNDED IN HEALTH EQUITY AND WITH CAREFUL ATTENTION TO SOCIAL AND STRUCTURAL DRIVERS OF HEALTH, WE WILL EXPAND THE SCOPE AND RELEVANCE OF SCIENTIFIC KNOWLEDGE AND TREATMENT INTERVENTIONS AT THE INTERSECTION OF THE HIV CARE CONTINUUM, THE ALCOHOL CARE CONTINUUM, AND HIV-RELATED COMORBIDITIES. WE WILL DO THIS BY DEVELOPING, TAILORING, AND IMPLEMENTING EVIDENCE- BASED INTERVENTIONS (EBIS) FOR ALCOHOL REDUCTION THAT ARE ACCESSIBLE TO PWH WITH UAU; ADDRESSING SYNDEMIC CONDITIONS IN OUR INTERVENTION, IMPLEMENTATION, AND EPIDEMIOLOGY WORK; ACTIVELY ENGAGING PWH WITH DIVERSE LIVED EXPERIENCES IN OUR CENTER; AND EXPANDING THE DIVERSITY AND INCLUSION OF PROJECT RESEARCHERS AND TRAINEES. OUR SCIENTIFIC GOALS ARE TO: 1) INCREASE THE REACH OF ALCOHOL EBIS THROUGH IMPLEMENTATION SCIENCE AND EPIDEMIOLOGICAL RESEARCH AT THE INTERSECTION OF UAU AND HIV, 2) EXPAND OUR HIV RESEARCH TO INCORPORATE NEW DEFINITIONS OF ALCOHOL USE DISORDER RECOVERY, AND 3) INCREASE INCLUSIVITY/DIVERSITY OF PERSPECTIVES IN ARCH REACH THROUGH COMMUNITY ENGAGEMENT AND MENTORING OF TRAINEES. WE PROPOSE A HIGHLY INTERACTIVE AND SYNERGISTIC CENTER WITH THREE PROJECTS SUPPORTED BY ADMINISTRATIVE, METHODS, AND DISSEMINATION CORES. ARCH REACH IS EMBEDDED IN AND CAPITALIZES ON THE RESEARCH AND CLINICAL INFRASTRUCTURE OF THE CENTER FOR AIDS RESEARCH NETWORK OF INTEGRATED CLINICAL SYSTEMS (CNICS), A MULTI-SITE CLINICAL COHORT REPRESENTING TEN UNIVERSITY-BASED HIV CLINICS ACROSS THE UNITED STATES (US)—WITH MANY LOCATED IN HIGH PRIORITY REGIONS FOR EHE. THE FIRST PROJECT LEVERAGES CNICS DATA TO EXAMINE THE RELATIONSHIP BETWEEN UAU, UAU TREATMENTS, AND NEW DEFINITIONS OF AUD RECOVERY ON HIV TREATMENT OUTCOMES AND COMORBIDITIES AMONG PWH IN THE CONTEXT OF SOCIAL DETERMINANTS OF HEALTH. OUR SECOND PROJECT, USING HUMAN CENTERED DESIGN METHODS, WILL ADAPT, TAILOR, AND PILOT TEST AN ALCOHOL EBI FOR TELEPHONE DELIVERY IN A HYBRID TYPE 1 EFFECTIVENESS-IMPLEMENTATION STUDY. LASTLY, OUR THIRD PROJECT, A HYBRID TYPE 2 EFFECTIVENESS-IMPLEMENTATION DESIGN, WILL TAILOR AND IMPLEMENT A SYNDEMIC CONDITIONS INTERVENTION FOR UAU AND CO-OCCURRING MENTAL HEALTH DISORDERS USING AN AUDIT AND FEEDBACK IMPLEMENTATION STRATEGY. AN ADMINISTRATIVE CORE WILL PROVIDE THE STRUCTURE AND PROCESSES TO SUPPORT COLLABORATION AND SYNERGY ACROSS ARCH REACH PROJECTS AND INVESTIGATORS. A METHODS CORE WILL SUPPORT IMPLEMENTATION SCIENCE AIMS OF PROJECTS AND DEVELOP NOVEL ANALYTIC METHODS. LASTLY, A DISSEMINATION CORE WILL ASSEMBLE A TRANSDISCIPLINARY COMMUNITY PARTNER BOARD AND SUPPORT THE DISSEMINATION OF PROJECT FINDINGS TO ENSURE THE EQUITABLE UPTAKE, RECEIPT, AND DELIVERY OF EBIS FOR UAU. CAPITALIZING ON A STRONG ORGANIZATIONAL STRUCTURE, THE EXTENSIVE RESOURCES AND INFRASTRUCTURE OF CNICS, AND A TRANSDISCIPLINARY TEAM INCLUSIVE OF COMMUNITY MEMBERS, ARCH REACH ADDRESSES ISSUES OF HIGH RELEVANCE TO US EHE GOALS BY ADDRESSING THE INTERSECTION OF HIV, ALCOHOL USE, AND HEALTH EQUITY.

GENOMIC AND PROTEOMIC DETERMINANTS OF RECURRENT VENOUS THROMBOEMBOLISM - ABSTRACT IN THOSE WHO SURVIVE THEIR INITIAL VENOUS THROMBOEMBOLISM (VTE) EVENT, EITHER A DEEP VEIN THROMBOSIS (DVT) OR A PULMONARY EMBOLISM (PE), RECURRENCES ARE COMMON: WITHIN 5 YEARS, 20-30% WILL HAVE EXPERIENCED A RECURRENT EVENT. ALTHOUGH A RECURRENT DVT CAN INFLICT LIFE-LONG VASCULAR MORBIDITY, A RECURRENT PE OCCURS IN 40% OF CASES AND IS FATAL IN 10%. PREVENTING THE RECURRENCE OF A VTE REMAINS A CRITICAL CHALLENGE FOR HEALTHCARE PROVIDERS AND THE PATIENTS THEY TREAT. ALTHOUGH WARFARIN AND THE NEWER DIRECT ORAL ANTICOAGULANTS (DOACS) ARE EXTREMELY EFFECTIVE AT REDUCING THE RISK OF VTE RECURRENCE, THEIR USE INCREASES THE RISK OF MAJOR, LIFE-THREATENING BLEEDS. RECENT EPIDEMIOLOGIC DATA SUGGEST THAT THE BIOLOGY UNDERLYING VTE RECURRENCE IS SIMILAR YET DISTINCT FROM INCIDENCE. IN THIS APPLICATION WE PRESENT EVIDENCE IN SUPPORT OF THIS DISTINCTION REGARDING GENETIC AND PROTEOMIC RISK FACTORS. THE LIMITED UNDERSTANDING OF VTE RECURRENCE RISK FACTORS AND PATHOBIOLOGY IS A SIGNIFICANT GAP IN CLINICAL AND PUBLIC HEALTH KNOWLEDGE AS WE HAVE FEW TOOLS TO IDENTIFY WHO IS AT GREATEST RISK, AND WE HAVE NO EFFECTIVE TREATMENT OPTIONS SAFER THAN DOACS. THIS GAP IS PARTICULARLY UNFORTUNATE SINCE THE RATES OF VTE RECURRENCE ARE HIGH AND THE AT-RISK POPULATION IS ALREADY UNDER CLINICAL CARE. WE PROPOSE TO ADDRESS KNOWLEDGE GAPS IN THE BIOLOGIC UNDERSTANDING OF VTE RECURRENCE BY INVESTIGATING GENOMIC AND PROTEOMIC DETERMINANTS IN INCEPTION COHORTS OF VTE. OUR GOAL IS TO IDENTIFY NEW BIOLOGY CONTRIBUTING TO THE RISK OF RECURRENT VTE. WE PROPOSE 2 AIMS THAT APPLY COMPLEMENTARY OMIC INTERROGATIONS TO RECURRENT VTE, AND WE HAVE PRELIMINARY DATA SUPPORTING OUR APPROACH. AIM 1: GENOMIC ASSOCIATIONS WITH RECURRENT VTE: USING AN INCEPTION COHORT OF ~16,000 VTE SURVIVORS WHO SELF-REPORT BLACK, HISPANIC, AND WHITE IDENTITIES IN THE DEPARTMENT OF VETERANS AFFAIRS MILLION VETERAN PROGRAM (MVP), WE WILL PERFORM A GWAS (USING EXISTING DENSELY IMPUTED GENOME-WIDE MARKERS) FOR THE TIME-TO-EVENT OF ~3,200 VTE RECURRENCES. REPLICATION OF STATISTICALLY SIGNIFICANT SIGNALS WILL BE CONDUCTED USING AN INDEPENDENT SAMPLE OF MVP AND EXTANT INVENT CONSORTIUM VTE GWAS SUMMARY DATA FROM 8,355 VTE SURVIVORS WITH 2,175 RECURRENCES. AIM 2: PLASMA PROTEIN ASSOCIATIONS WITH RECURRENT VTE: FROM AN INCEPTION COHORT OF 1,961 VTE SURVIVORS IN THE SEATTLE HEART AND VASCULAR HEALTH VTE STUDY WITH BLOOD PLASMA COLLECTED AND STORED POST-INCIDENT EVENT, WE WILL TEST ASSOCIATION OF 5,400 VALIDATED PLASMA PROTEINS WITH VTE RECURRENCE USING A NESTED CASE-COHORT DESIGN OF 900 INCIDENT VTE SURVIVORS AMONG WHOM 450 HAD A RECURRENCE. WE CONDUCT REPLICATION IN 2,000 MULTI-ANCESTRY VTE SURVIVORS IN MVP, AMONG WHOM 225 EXPERIENCED A RECURRENT EVENT. PROTEINS WILL BE VALIDATED WITH MASS SPECTROMETRY. NEXT, WE WILL USE EXTANT AND NEW PROTEIN QUANTITATIVE-TRAIT LOCI RESOURCES AS GENETIC INSTRUMENTS IN MENDELIAN RANDOMIZATION WORK, COMBINING THEM WITH EXTANT GENOME-WIDE ASSOCIATION RESOURCES OF VTE RECURRENCES FROM AIM 1 TO EVALUATE CAUSAL EFFECTS OF THE PLASMA PROTEINS ON VTE RECURRENCE.

HOWEVER, THE INTERNATIONAL CONSENSUS STANDARDS (IEC 62600-40) ARE ONLY CAPABLE OF QUANTIFYING, IN A CONSISTENT MANNER, RECEIVED LEVELS IN THE VICINITY OF WEC. THIS DOES NOT ALLOW TECHNOLOGY DEVELOPERS OR ENVIRONMENTAL REGULATORS TO DEFINITIVELY ATTRIBUTE RADIATED NOISE TO A WEC OR UNDERSTAND HOW THESE RECEIVED LEVELS MIGHT CHANGE WITH OPERATIONAL (E.G., CONTROL STRATEGY) OR ENVIRONMENTAL (E.G., SEA STATE) COVARIATES. THE PROJECT OBJECTIVE IS TO DEVELOP A RELIABLE AND SCALABLE MANUFACTURING PROCESS (CALLED AFIELD) THAT USES ACOUSTIC FIELD-ASSISTED ASSEMBLY OF PARTICLE-BASED MATERIALS FOR NEXT-GENERATION STRUCTURED ELECTRODES (SES) FOR HIGH-ENERGY, HIGH-POWER, FAST CHARGE LI-ION BATTERY PACKS. AFIELD USES ACOUSTIC WAVES, AND THESE WAVES CREATE AN ACOUSTIC FIELD THAT CAN BE USED TO ASSEMBLE PARTICLES IN A FLUID ON A MICRON-SCALE. THIS PROJECT IS DIVIDED INTO THREE BUDGET PERIODS (BPS) WITH DISTINCT OBJECTIVES TO PUSH THE TRL OF THIS TECHNOLOGY FROM 2 TO 4 BY PROJECTION COMPLETION. BP 1 FOCUSES ON SCALING UP AFIELD FROM A MM-SCALE TO CM-SCALE PROCESSING SYSTEM WHILE DEFINING OPTIMAL SES FOR FABRICATION. BP2 ENTAILS REFINING AND FURTHER SCALING UP THE INITIAL AFIELD PROTOTYPE AND VALIDATING IT FOR SE BATTERY FABRICATION WHILE TESTING AN IN-SITU METROLOGY AND INSPECTION WORKFLOW TO IMPROVE PROCESS RELIABILITY. LASTLY, BP 3 CONCLUDES THE PROJECT WITH A LARGE SCALE (= 10 AH) MODELING ANALYSIS OF THE THERMAL AND ELECTROCHEMICAL BEHAVIOR OF AFIELD SES WHILE OPTIMIZATION AND FURTHER SCALE-UP OF THE AFIELD CHAMBER FOR RELIABLE SE PROCESSING IS CONDUCTED WITH MORE DETAILED MICROSTRUCTURAL AND ELECTROCHEMICAL CHARACTERIZATION. THROUGH BPS 1-3, THIS PROJECT WILL RESULT IN FOUR KEY OUTCOMES: (1) SUCCESSFUL APPLICATION OF ACOUSTIC FIELDS TO FABRICATE CM-AREA SE LI-ION BATTERIES (2) FUNCTIONAL CATHODES/ANODES THAT DEMONSTRATE = 10% (MINIMUM) IMPROVEMENT IN CAPACITY OR CHARGE BEHAVIOR OVER A CONVENTIONAL BATTERY CELL, (3) IN SITU AND POST SITU METEOROLOGY AND INSPECTION WORKFLOWS FOR ENSURING RELIABLE SE FABRICATION, AND (4) AN IN-DEPTH PACK-SCALE ANALYSIS OF THE BENEFIT OF SES FOR ELECTRIC VEHICLES (EVS). THIS NOVEL AND DISRUPTIVE AFIELD PROCESS IS AN EXAMPLE OF THE MANUFACTURING INNOVATION NEEDED FOR BATTERY ENERGY STORAGE IN THE U.S. BECAUSE IT WILL ENABLE LARGE-AREA, CONSISTENT, CONTROLLABLE, AND ROBUST MANUFACTURING.
